Abstract:Todevelopanoninvasivemedium-basedpreimplantationgeneticdiagnosis(PGD)testfora-thalassemias-SEA.
Theembryosofa-thalassemia-SEAcarriersundergoinginvitrofertilization(IVF)werecultured.Singlecellswerebiopsiedfromblastomeresandsubjectedtofluorescentgappolymerasechainreaction(PCR)analysis;thespentculturemediathatcontainedembryogenomicDNAandcorrespondingblastocystsasverificationweresubjectedtoquantitative-PCR(Q-PCR)detectionofa-thalassemia-SEA.Thediagnosisefficiencyandalleledropout(ADO)ratiowerecalculated,andthecell-freeDNAconcentrationwasquantitativelyassessedintheculturemedium.
